Nitto is proud to announce the publication of our latest research on antibody-functionalized lipid nanoparticles (LNPs) in Nano Letters.
The paper, titled “Balancing Multivalent Avidity and Receptor Availability Governs mRNA Delivery by Antibody-Functionalized Lipid Nanoparticles,” presents a quantitative design framework for targeted mRNA delivery using antibody-functionalized lipid nanoparticles (LNPs).
The study reveals how surface avidity—the density of functional antibodies on the nanoparticle surface—critically determines delivery efficiency, exhibiting a bell-shaped relationship driven by the balance between receptor engagement and receptor preservation. By tuning ligand density, it achieved efficient and selective in vivo delivery to human CD8+ cells, enabling potent in vivo CAR-T generation at low doses.
This approach offers a practical design principle for advancing targeted mRNA therapeutics.
